摘 要:分别用碳源、氮源、铜离子、诱导剂等不同的培养方式优化液体培养条件,并研究其对灵芝TR6号菌株漆酶分泌能力的影响。结果表明:与静止培养相比较,摇床培养能明显促进灵芝TR6号漆酶的分泌。以可溶性淀粉(25g/L)为碳源,胰蛋白胨为氮源(2g/L),培养7d后添加硫酸铜使铜,使离子浓度达到1.0mmol/L。以0.03mmol/LABTS作诱导剂,灵芝TR6号菌株分泌漆酶最多,漆酶活性可达11000U/L。In this paper, some different culture conditions such as carbon source, nitrogen source, copper ion concentration, and inducer were optimized and studied on the influence of liquid culture conditions on production capacity of Ganoderma Ineidum strain TR6 laccase. Results indicated that shake-bed culture with 110 r/min could accelerate laccase production significantly compared to the stationary culture. The maximum laccase activity of 11 000 U/L was achieved under the conditions of 25 g/L of soluble starch as carbon source, 2.0 g/L of tryptone as nitrogen source, 1.0 mmol/L of copper ion concentration as metal additive(adding it after culture 7 d),0.03 mmol/L ABTS as inducer.
