摘 要:采用PEG/磷酸钾盐双水相系统,从离心除菌后的发酵液中提取聚谷氨酸。研究PEG分子量、PEG浓度、磷酸钾盐浓度、磷酸钾盐缓冲液的pH值、发酵液加量和环境温度对聚谷氨酸分配系数和提取率的影响。正交试验结果显示聚谷氨酸富集于上相,当系统组成为24%(质量分数)PEG1500,23%(质量分数)磷酸钾盐,磷酸盐缓冲液pH值为8.0,发酵液的加入量为10%(质量分数)时,聚谷氨酸的分配系数达到31.7,上相收率为96.8%以上,自然分相时间小于5min。研究结果表明,采用PEG/磷酸钾盐双水相系统,从离心除菌后的发酵液中提取聚谷氨酸是可行的。The work attempt to study and optimize the aqueous two-phase systems (PEG/potassium phosphate) to extract γ-PGA from fermentation broth,in which the cell was removed by centrifugation,Influence of several parameters such as phase forming polymer molecular weight,concentration of phase forming components (polyethylene glycol,potassium phosphate,fermentation broth),pH of the potassium phosphate buffer solution and tempera- ture on differential partitioning of γ-PGA required for the extraction ratio and partition coefficient were studied. The results from orthogonal test showed that T-PGA was enriched to the top phase. The constituted 24 %(w/w) PEG1500 with 23 %(w/w) potassium phosphate by the aqueous two phase system with 10 %(w/w) fermentation broth at the conditions of pH=8.0 of the potassium phosphate buffer solution.The partition coefficients obtained as 31.7 and the extraction ratio was 96.8 %,partitioning time was less than 5 min,Thus the aqueous two-phase system (PEG/potassium phosphate) provide a new method for separation of γ-PGA.
