摘  要:以新霉素的降解产物新霉胺作为半抗原,采用戊二醛法合成新霉胺免疫原,制备出针对多种氨基糖苷类药物的多克隆抗体。使用高碘酸盐法制备新霉胺的酶标抗原,建立氨基糖苷类药物的直接竞争酶联免疫方法。结果表明:该方法针对新霉素、庆大霉素和卡那霉素的检出限分别为0.2、0.15和0.35mg/kg,低于我国制定的最高残留限量0.5mg/kg。此方法灵敏、准确,适用于氨基糖苷类药物多残留免疫检测的要求。For the immunization of rabbits, neamine was used as the generic hapten on the basis of the facts that it is a degradation product of neomycin. Neamine was linked to the carrier protein keyhole limpet hemocyanin by GA coupling. The immunogen of Neamine was synthesized by glularaldehyde method and the polyclonal antibodies were prepared. The enzyme antigen of neamine was prepared by periodate method. The specificity and sensitivity of the polyclonal antibodies for the aminoglycoside antibiotics were tested in a competitive direct ELISA. Neomcyin, gentamicin, and kanamycin were detected in PBS with 15% inhibition levels at 0.2,0.15,0.35 mg/kg, respectively. Levels below the China maximum residue levels (0.5 mg/kg) for aminoglycoside antibiotics could readily be measured in this generic ELISA. This sensitive and accurate method could be used for the simultaneous determination of aminoglycoside antibiotics.
