摘  要:从江苏连云港海域筛选和分离到1株产低温淀粉酶的海洋细菌Pseudoalteromonas sp.G23,对该菌株进行了产α-淀粉酶发酵条件研究,结果表明,该菌株发酵16h后到达产酶高峰,最适产酶温度为15℃,最适产酶pH值为8.5,装液量为20%。可溶性淀粉、酵母膏和蛋白胨促进产酶。利用响应面方法对菌株Pseudoalteromonas sp.G23产α-淀粉酶的发酵条件进行了优化,结果表明,可溶性淀粉浓度为1.16%、蛋白胨浓度为2.11%,发酵温度12.38℃,酶活为41.4U/mL,较优化前提高了7倍。A cold-active amylase producing marine bacterium Pseudoalteromonas sp. G23 was screened and isolated from the sea area of Lianyungang, Jiangsu. The production of α-amylase from this strain has been studied. Maximum enzyme production was achieved after 16h cultivation. The optimum temperature, pH and medium volumn were 15℃, 8.5 and 20%, respectively. The addition of soluble starch, yeast extract and peptone enhanced enzyme production. The response surface design was used to optimize the fermentation conditions for α-amylase production by Pseudoa teromonas sp. G23. The optimum conditions were soluble starch 1.16%, peptone 2.11%, and fermentation temperature 12.38℃. After optimizatiol the amylase activity was 41.4 U/ml, which was increased by 7 times compared with that before optimization.
