摘 要:为了获得较高的多糖提取率,采用超声波协同纤维素酶法提取霍山石斛多糖;研究了料液比、纤维素酶用量、酶解温度、酶解时间、超声波功率和超声时间等因素对多糖提取的影响,同时通过正交实验对其提取条件进行了优化。结果表明,最佳提取工艺条件为:料液比1∶40(g/mL)、纤维素酶500U/g、酶解温度40℃,酶解时间3h,超声波功率为300W,超声时间6min,此时的多糖提取量达到0.283g/g。In order to obtain preferable extraction rate of polysaccharides from Dedrobium huoshanense,the effects of the ratio of material to water, cellulase dosage, reacting temperature and time, ultrasonic operative power and time on the extraction of polysaccharides were investigated by ultrasonic and cellulase method and the preferable conditions for extracting polysaccharides were obtained by orthogonal experimental method.The results showed that the optimal conditions for extracting polysaccharides were as follows: the ratio of material to water 1:40 ( g/mL), cellulase dosage 500U/g, reacting temperature 40℃, reacting time 3 h, ultrasonic operative power 300W, and ultrasonic operative time 6min and the extraction rate of polysaccharide reached 0.283g/g.
