摘  要:用水提法提取沙苑子多糖,并对其脱蛋白方法进行了比较研究。结果表明,水提沙苑子多糖的最佳条件是:提取温度为65℃,提取时间为3h,料液比为1∶15,提取次数为3次。在该提取条件下,可以得到最高粗多糖得率96.9g/kg;分别采用sevag法、酶法、三氯乙酸(TCA)法和sevag法结合酶法等方法脱去沙苑子粗多糖提取物中的蛋白质。酶法脱蛋白的最佳条件为:温度为45℃,时间为1h,酶量为0.4g,pH值为7.0。结果表明,用sevag法结合酶法脱蛋白的效率最高,为87.5%,且多糖损失率较低,为14.9%。The conditions for extracting polysaccharides with water from Flatstem milkvetch seed, including incubation time and temperature, times of extraction and feed liquid ratio, were investigated through an orthogonal design of L(34)in this work.. The highest yield of 96.9 g/kg was achieved by in the water bath at 65 ℃ for 3 h with the feed liquid ratio at 1 : 15, then followed the same process three times. Four methods were used to remove the protein from polysaccharides, and the results showed the combination enzyme and Scrag exhibited higher removal percentage of protein and lower loss of polysaccharide. The proper weight of papain is 0.4 g, the temperature is 45℃, time for reaction is 1 h, pH is 7.0, and the times of Sevag to remove protein is 3. After the optium conditions, the removal percentage of protein is 87.5%, and the loss of polysaccharides is 14.9%.
