EFFECTS OF NITROGEN ON TISSUE CULTURE AND AGAVAIN-SH INDUCEMENT OF Agave sisalana

摘  要:以LS培养基,附加特定种类和浓度的氮素,再加入2,4-D40mg/L,NAA04mg/L,6-BA25mg/L,比较了不同氮素类型,不同氮源浓度对剑麻愈伤组织细胞生长速率及蛋白酶活力的影响结果表明,(1)同时含有铵态氮和硝态氮的无机氮源培养基最有利于剑麻愈伤组织细胞的快速增长;(2)1~10g/L的有机氮源有利于提高剑麻蛋白酶的活力;(3)剑麻愈伤组织细胞生长速率与蛋白酶活力呈相斥型生长关系,因此在剑麻蛋白酶的组培诱导中,以采取二步培养法为宜.allus as cultured in LS medium added with 2,4-D 3 mg/L,NAA 0.4 mg/L,6-BA 2.5 mg/L,and effects of various kinds and concentrations of nitrogen on its growth rate and special activity of agavain-SH were studied.The results showed that the medium containing both ammonium and nitrate as nitrogen nutrition was the best for the fast growth of the callus,that the medium containing 1~10 g/L of organic nitrogen was effective for improving the special activity of agavain-SH, and that the callus growth rate of Agane sisalana was negatively correlated with the special activity of agavain-SH.For these reasons the tissue culture and agavain-SH inducement of Agave sisalana  should be better to performed by two-stage culture method.
