摘  要:对含有rhNGF基因的工程菌EscherichiacoliDHI/pJN的发酵条件进行了较详细的研究,探讨了培养基组成,培养时间、诱导时间以及补加葡萄糖对菌体生长和产物表达的影响,确定了该工程菌发酵的最佳条件。The culture conditions of an engineering bacteria Escherichia coliDHI/pJN expressed rhNGF have been investigated. The results from the relationship between the growth condition of the strain including media, culture time, inducing time, feeding glucose,etc and the expression product of target protein showed that the complex media BM+0.5%YE could obviously increase the growth of cells and the expression product. The experiment also indicated that the yield of rhNGF could reach a higher level at the middle of logarithm phage,30℃, and raise the temperature at 42℃, 4h for inducing, As well as, during the periods of culture, glucose was fed to make its concentration at the level of ca. 0.1% so that cells could absorb more nutrient and increase its density.
