摘 要:为进一步提高γ-聚谷氨酸(γ-polyglutamic acid,缩写γ-PGA)成品质量和提取效率,采用活性炭脱色和硅藻土吸附-抽滤替代离心的预处理方法,有效地实现了菌液分离以及色素和杂蛋白的去除。发酵液中加入硅藻土和活性炭的最适用量分别为20g/L和0.5g/L,粗细硅藻土的最佳配比为1.5:1,抽滤的最佳pH条件为pH3.0~3.2。预处理后的发酵液依次经过超滤浓缩和醇析的纯化过程,最终真空干燥得到氢型或钠盐型的γ-PGA成品,总收率达90.6%,产品为白色无定型粉末,质量指标已达到化妆品级的要求。A novel pretreatment method was developed to separate and purify γ-PGA from culture broth. The viscosity of culture broth was decreased by adjusting solution pH to acidic condition using trichloracetic acid. Diatomite and active carbon was added as absorbent and decoloring agent respectively. Then, the solution was filtrated in vacuum with diatomite to remove cells, pigments and proteins. The optimum quantity of using mixed diatomite loading (ratio of coarse to small diatomite 1.5 : 1) and active carbon added in culture broth was 20g/L and 0.05g/L respectively, and the optimal pH during absorption and filtration was controlled at 3.0-3.2. The ultrafiltration membrane was used to deionize and condense γ-PGA. Purified γ-PGA was then obtained through precipitation by ethanol and drying in vacuum. The total yield of γ-PGA reached up to 90. 6%. The physical appearance of the prepared γ-PGA product was white free-flow powder and the quality parameter of it had achieved the cosmetic grade.
