摘 要:以橄榄油为唯一碳源,在含维多利亚兰的培养基上筛选来自油菜地土壤的样品。筛选到1株活力较高的菌种,经鉴定为成团肠杆菌(Enterobacter agglomerans),在优化的产酶条件(2.0%的乳糖作为碳源,1.5%的牛肉浸膏和1.0%的酵母膏作为复合氮源,0.1%MgSO4.7H2O,初始pH7.0,30℃)下培养48h,酶活为39.09U/mL。在反应温度为30℃,振荡速率为180r/min时催化能力较强。其粗发酵液可催化含水量在91%以上的橄榄油、芝麻油、茶油生产生物柴油。This paper screened rapeseed soil sample in Victoria blue solution solid medium with olive oil as sole carbon source. The lipase of Enterobacter agglomerans with the highest activity was identified. Under the optimum conditions the enzyme activity of this strain reached to 39.09 U/mL, and the conditions were. initial pH 7.0, agitating at 30 ℃ for 48 h, 2.0% lactose as carbon sources, 1.5% beef extract and 1.0% yeast extract as nitrogen source, 0.1% MgSO4 · 7H2O. Under the conditions of 30 ℃ and 180 r/min, a better result of the catalyzing reaction was observed. Olive oil, sesame oil and tea oil with more than 91% water can be catalyzed by the lipase of this strain to produce bio-diesel. We can produce bio-diesel from wasted oils using this strain, which can be an additional source of raw materials for bio-diesel and helping decrease the cost of bio-diesel production. Further research on this strain is worthwhile.
