摘 要:采用石油醚、乙醚、乙酸乙酯、正丁醇、乙醇、水6种溶剂分别提取巨大口蘑子实体中的各种不同成分;用牛津杯法进行抑菌实验,结果显示:乙醚提取物对受试菌的抑制作用较乙酸乙酯提取物强,其余提取物对受试菌没有直接的抑制作用;2mg/mL的乙醚提取物处理各受试菌,抑菌圈直径分别为:沙门氏菌(Salmonella)19·83mm,大肠杆菌(Escherichia coli)15·50mm,苏云金芽孢杆菌(Bacillus thuringiensis)14·83mm,金黄色葡萄球菌(Staphylococcus aureus)13·17mm,枯草芽孢杆菌(Bacillus subtilis)8·50mm,与乙醚对照相比差异显著;3mg/mL乙醚提取物对S·aureus、B·thuringiensis和E·coli的抑菌圈直径与0·1%新洁尔灭处理的抑菌圈直径差异不显著,对B·subtilis抑菌圈直径与0·1%HgCl2处理的差异不显著。最后用GC-MS对乙醚提取物的化学成分进行研究。HP-1柱分离出30个峰,用质谱法鉴定出20种成分,主要由酸类(61·15%)、酯类(28·01%)、烷烃(0·52%)、酮类(0·26%)等组成。The antimicrobial activity of the extracts of six different solvents namely petroleumether, aether, ethyl acetate, n-butanol,ethanol and water of Macrocybe gigantean(Massee) by the Oxford plates were studied and the main antimicrobial components of aether extract of Macrocybe gigantean(Massee) was analyzed by the GC-MS. The result showed that the inhibitory effect was good with the aether extract compared with the ethyl acetate extract and the other extracts were no inhibitory effect to the tested bacteria; the antibacterial circle diameters with the treatments of 2mg/mL aether extract were Salmonella 19.83mm, Escherichia colt 15.50mm, Bacillus thuringiensis 14.83mm, Staphylococcus aureus 13.11mm and Bacillus subtilis 8.50mm respectively and were significantly different compared with the control.No obvious difference was found in the bacterial inhibition diameters of Soaureus, B.thuringiensis and E.cofi with the 3mg/mL aether extract compared with 0.1% benzalkonium bromide,and the bacterial inhibition diameters of B.subtilis had not significant difference between the aether extract and 0.1 % mercuric chloride.The chemical compositions of the ether extract have been identified by means of the Gas Chromatography-Mass Spectrometry, Twenty compositions have been identified from 30 separated peaks.The major compositions are acids (61.15%), (0.26%) etc.
