摘  要:对VB1、L-谷氨酸、柠檬酸和油酸等四种生长因子对裂褶菌胞外多糖产量的影响进行了优化研究。研究发现,四种因子可不同程度地促进裂褶菌菌丝生长和其胞外多糖的生成。通过单因子实验,初步确定了各种生长因子的适宜添加浓度。采用VB1、L-谷氨酸、柠檬酸和油酸为四个实验因素,进行了L16(45)正交实验,四种因子对裂褶多糖产量影响的显著顺序为:油酸>柠檬酸>VB1>L-谷氨酸;获得四种因子的优选水平组合:VB10·5mg/L、L-谷氨酸0·5mg/L、柠檬酸0·05g/L、油酸0·2mg/L;在该优化条件下进行裂褶菌培养,其胞外多糖产量达到5·73g/L,比空白对照样提高了63·71%(P<0·05)。The effect of four growth factors on the production of exopolysaccharide production by Schizophyllum commune was studied.All of them had a positive effect on cultivation of So commune.The one-factor-at-a-time method was adopted to investigate the effects of four different compounds( VB, L-glutamate, citric acid, and oleic acid) on mycelial growth and exopolysaccharide production.All these factors were identified to be the most suitable factors.Subsequently,the concentrations of four growth factors were optimized using the orthogonal matrix method.The effects of the factors on the schizophyllan production of S.commune were in the order of oleic acid 〉 citric acid 〉VB, 〉 L-glutamate.The optimal concentration for enhance production were determined as VB, 0.5mg/L, L-glutamate 0.5mg/L, citric acid 0.05g/L and oleic acid 0.2mg/L for schizophyllan production.This optimization strategy in shake flask culture lead to a schizophyllan production of 5.73g/L, and it had 63.71% more than blank sample( P 〈0.05).
