摘 要:目的:建立测定马铃薯叶中茄尼醇的反相高效液相色谱(RP-HPLC)分析方法。方法:采用Agilent 1100 HPLC,XDB-C18色谱柱(250mm×4·6mm,5μm),流动相V(甲醇)∶V(乙醇)=45∶55等度洗脱,洗脱流速1·0mL/min,柱温35℃,检测波长211nm,保留时间约8·750min。结果;茄尼醇质量浓度在6·125~196μg/mL(R=0·9999)范围内线性关系良好,平均加样回收率(n=9)为99·04%,RSD=1·87%。通过与传统提取方法比较,确定最佳的提取方法为微波辐射法。结论;该方法简便、快速、准确、重复性好,适用于马铃薯叶中茄尼醇的含量测定。Objective: A sensitive RP-HPLC method was established for determining the content of solanesol in the extracts of potato leaves.Methods :The Agilent 1 100 HPLC system,A I.D.250mm × 4.6mm, particle size 5μm Agilent XDB-C18 column,was operated at a column temperature of 35℃.The mobile phase was V( carbinol) :V( ethanol) = 45:55 at a flow rate of 1.0mL/min.211 nm was set as the UV detection wavelength for solanesol, retention time was probable 8.750min.Results: The experimental results showed a good linear relationship within 6.125 - 196μg/mL ( R =0.9999) ,the average recoveries( n = 9) were 99.04%, RSD = 1.87/%.Compared with the conventional methods of extraction, the optimum extraction method was Microwave-assisted extraction.Conclusion: The method was rapid,simple,accurate and good reproducibility, it was suitable for the determination of solanesol in the extracts of potato leaves.
