摘  要:目的探讨产胞外蛋白酶细菌的快速筛选方法和产酶条件优化。方法取昆明郊区的农田土壤及河沟淤泥,采用含脱脂奶粉的菌种筛选培养基和划线法分离产蛋白酶的菌株,选用5种培养基进行发酵产酶条件优化。结果农田土壤中含有种类较多的产蛋白酶的菌株,菌种单一性不强。YPD培养基发酵的菌种蛋白酶活性高于其他4种培养基。结论利用含脱脂奶粉的筛选培养基进行初筛,YPD进行发酵培养复筛,可以显著提高筛选效率。Objective : To discuss the method of rapid screening technique for extracellular protease-generating bacteria and the best conditions for protease fermentation. Methods: The protease producing strains were isolated from farmland soil and sludge, with skim milk isolating medium and streak culture. The conditions were optimized by using five kinds of fer- mentative medium for protease fermentation. Results: The sample from farmland had more species of protease producing strains. Various shapes of bacteria were observed by optical microscope. The bacteria had higher protease activity after fermentation by the use of YPD liquid medium than those by the use of other four kinds of mediums. Conclusion : The efficiency of isolation is raised by the use of the skim milk isolating medium and YPD liquid medium in the isolation of protease producing bacteria.
