摘  要:采用DTF-02型阳离子交换树脂对酶法合成的双果糖酐Ⅲ进行了分离纯化,探讨了柱温、进样浓度、进样体积和流速对分离效果的影响。结果表明:选择柱温60℃、流速1 mL/min、进样体积5 mL、固形物含量20%的条件下,可以得到完全分开的双果糖酐Ⅲ和低聚果糖,此时收集含DFAⅢ的洗脱液,DFAⅢ含量在95%以上。The separation and purification of difructose anhydride prepared by enzymatic synthesis using cation exchange resin DTF-02 was studied. The influence of column temperature, sample concentration, sample volume and elution flow rate were discussed. The study showed that, when 5mL difruetose anhydride Ⅲ containing a soluble solid content of 20% , difructose anhydrideⅢ was completely separated from fructo-oligosaccharide with elution flow rate of 1 mL/min at 60℃. Under such condition, the fractions containing DFAⅢ were separated and determined by HPLC which showed DFA Ⅲ content was over 95%. The research laid theoretical foundations for large-scale separation and purification of difructose anhydride Ⅲ.
