摘 要:目的:研究一种基于近红外(NIR)透射光谱法的甜菊糖有效成分快速测定新方法。方法:采用NIRS分析技术,对不同批次的甜菊糖苷水溶液采集透射光谱,选取不同的预处理方法和波段范围对光谱数据进行有效的信息提取与分析,确定最佳的回归因子数以及用于定量分析的最优波段范围。采用偏最小二乘法(PLS)建立甜菊糖苷水溶液中有效成分含量的定量分析模型。结果:甜菊糖溶液中甜菊双糖甙(steviolbioside,SX)、杜克甙A(dulcoside A,DA)、甜菊糖甙(stevioside,STV)、瑞鲍迪甙C(rebaudioside C,RC)、甜菊糖A3甙(Rebaudioside A,A3)和甜菊糖总苷(steviosides,STS)的PLS模型相关系数(R2)分别为0.9271、0.9355、0.9759、0.9706、0.9500、0.9870,预测均方根误差(RMSEP)分别为0.0004、0.0015、0.0150、0.0043、0.0170、0.0199。结论:所建方法操作简便、快速、准确、无损,具有较好的定性和定量分析性能,可用于甜菊糖苷溶液中各有效成分的快速检测。Objective:To establish a new method of assaying rapidly of active components in steviol glycoside solution. Method: The near infrared transmittance spectroscopy was used to quantify the different batches of steviol glycoside. According to selected different preprocessions and wave bands for information extraction and analysis from spectrums, the best factors and wave bands for quantitative analysis were determined. The calibration models were established using the PLS (partial least squares). Result: The correlation coefficients (R^2) for steviolbioside(SX), dulcoside A(DA), stevioside (STV), rebaudioside C(RC), Rebaudioside A(A3) and steviosides(STS) were 0.9271, 0.9355, 0.9759, 0.9706, 0.9500 and 0.9870, and the RMSEP were 0.0004, 0.0015, 0.0150, 0.0043, 0.0170 and 0.0199, respectively. Conclusion: The proposed method was simple and fast, and it provides a novel efficient and environmental friendly approach for the fast determination of the active components in steviol glycoside.
