摘 要:采用荧光光谱法测定沙米中精氨酸含量,对荧光光谱仪工作条件和标准曲线制作的最优条件进行了研究。结果表明:荧光分析的最佳条件是,发射波长是473nm,精密度S=3,纵轴扩大值Y=1;制作标准曲线的测定条件为是,用pH=6.0的乙酸-乙酸钠缓冲体系,试剂加入顺序依次为精氨酸(或精氨酸样品)、乙酰丙酮、甲醛、缓冲溶液;在12h内测定;精氨酸浓度与荧光强度在1-11μg/mL内呈直线关系,最低检测浓度1μg/mL;平均加标回收率达到98.55%。将该法用于样品分析,沙米中精氨酸含量达2.58%,结果表明,荧光光谱法适合于沙米中精氨酸含量的测定。The mal conditions were mission wavelength L-arginine was determined by fluorescence spectrophometry in Agriophyllum quarrosum. The optitested. The results showed that the optimum conditions of fluorescence spectrophometry were : e- 473nm, sensory 3, Y at 1. The testing conditions of making standard curve were pH 6.0 in the acetic acid and sodium acetate buffer solution. The order of the reagents added was arginine(or sample) , acetylacetone, formaldehyde and buffer. Arginine must be measured within 12h. The linear relationship existed between the arginine concentration and fluorescence intensity in the range of 1.0 -20.0μg/mL of arginine. The lowest concentration for detecting was 1.0μg/mL. Average sample recovery rate was 98.55% o The arginine content in Agriophyllum quarrosum was up to 2.58% determined under the optimal condition. Therefore the fluorescence spectrophotometric method could be used to determine arginine in Agriophyllum quarrosum.
