摘 要:用硅藻土对实验室筛选得到的成团肠杆菌脂肪酶干燥酶粉进行固定化,固定化酶在有机溶剂体系下催化生产生物柴油。在最佳反应条件,即菜籽油15.47 mL,固定化脂肪酶用量1 000 U,甲醇为酰基受体(7.15 mL,3次等量加入),5 mL正己烷,振荡速度180 r/min,35℃反应48 h时,转化率达91.03%。实验结果表明,油酸含量高有利于生产生物柴油,而芥酸有不利影响。固定化酶稳定性好,重复使用8次,转化率仍大于50%,同时还具有一定的适应性,可催化大豆油和葵花籽油生产生物柴油。研究表明,固定化酶可用于催化生产生物柴油,并有效降低酶催化法的生产成本。The lipase of Enterobacter agglomerans was immobilized by diatomite, and used to catalyze the production of biodiesel in a solvent system. The best reaction rapeseed oil 15.47 mL, dosage of immobilized lipase 1 000 methanol, equivalent adding for 3 times), 5 mL n -hexane, conditions were as follows: the volume of U, methanol as acyl acceptor (7. 15 mL oscillation speed 180 r/min, reaction ternperature 35 ℃, time 48 h. Under the best conditions, the conversion rate was 91.03%. It found that higher oleic acid content in the raw materials could improve the transesterification rate, while higher erucic acid content could lower the transesterification rate. The conversion rate was still more than 50%, even when the immobilized lipase was used for 8 times, and the lipase could catalyze soybean oil and sunflower seed oil to produce biodiesel with a better conversion rate. The results showed that the lipase of Enterobacter agglomerans could effectively reduce the cost of biodiesel produced by enzyme - catalysis, and it was a promising lipase for biodiesel production.
