摘  要:从富含蛋白质的土壤中筛选出的碱性蛋白酶产生菌,命名为Bacilussp.8(简称为No.8)。No.8菌能在pH6-12且含有较高浓度钙离子条件下生长并产酶,与目前的碱性蛋白酶生产菌如地衣芽孢杆菌2709相比,它具有较强的耐碱性和耐钙离子性。培养基起始pH值为7.5,接种菌龄为18h种子液2%。在35~37℃,培养48~60h的条件下,No菌产生的蛋白酶活力可稳定在1700~1800u/ml,并且在浸灰过程中可以脱毛。酶的最适pH为9.6,pH10.6-11.6最稳定。最适温度为40℃。A alkaline unhairing protease producing strain, named Bacillus sp.8(simply No.8) has been isolated from rich protein-containing soil for the first time. Fermentation condition of the strain has been studied. The results showed that soy bean cake meal and sweet potato meal(maize meal or glucose) are the best nitrogen and carbon sources for the isolated strain to produce proteases. The optimum conditions for enzyme production were:initial pH7.5, 35-37℃for 48-60h. 0.4-0.6% Na 2HPO 4 had a positive effect on the enzyme production. Strain No8 produced maximum alkaline protease activity 1800u/ml. The optimum pH and temperature for the protease activity production are pH 9.6,40℃. The enzyme were stable over pH 10.6. Comparing with the alkaline producing producer B.licheniformis 2709, the isolated strain has higher enduring of alkaline and Ca 2+ .
