摘 要:目的:研究大蒜生理活性物质对人胃癌细胞MGC-9和小鼠黑色素瘤细胞B16细胞增殖及细胞周期的影响。方法:用倒置显微镜观察细胞形态,噻唑蓝(MTT)比色法测定细胞增殖抑制率;流式细胞仪检测B16细胞周期分布的影响。结果:经大蒜素及其前体物质蒜氨酸、蒜氨酸酶和蒜氨酸+蒜氨酸酶处理后,MGC-9和B16细胞不同程度变圆,体积缩小,脱壁细胞增多;除蒜氨酸酶外,蒜氨酸、大蒜素和蒜氨酸+蒜氨酸酶都能诱导MGC-9和B16产生时间和剂量依赖性的增殖抑制作用,其中蒜氨酸+蒜氨酸酶的作用尤为显著。大蒜素和蒜氨酸+蒜氨酸酶阻滞B16细胞于G0/G1期。结论:本文由大蒜提取物处理后结果与已发表的临床研究相一致。AIM: To study the effect of garlic extracts on cell proliferation and cell cycle on human gastric cancer cell line(MGC-9) and mouse murine melanoma cell line(B16).METHODS: Cell morphology was ob-served by inverse microscopy.Cytotoxicity was examined by dimethyl thiazolyl tetrazolium bromide assay.Cell cycle distribution was detected by flow cytometry.RESULTS: After being treated with allicin and its precursors-alliin,alliinase and mixture of alliin and alliinase from garlic extracts,cells rounded up,shrank and cell membrane lost its integrity.Allicin,alliin and mixture of alliin and alliinase were all able to exert antiproliferative effects on MGC-9 and B16 cells in a dose-and time-dependent manner,and the inhibitory potency of mixture of alliin and alliinase was more than that of others.Allicin and mixture of alliin and alliinase could arrest the cell cycle of B16 in G0 /G1 phase.CONCLUSION: In experiment the results given by a treatment of garlic extracts are in good agreement with observation established in clinical studies.
