摘  要:采用离子交换色谱对发菜多糖进行分离纯化,对OH-型和Cl-型交换色谱对发菜多糖分离纯化效果进行初步的比较。离子交换色谱柱(Ф2.6×30),采用DEAE-Cellulose离子交换树脂和NaCl梯度洗脱对发菜多糖进行纯化。使用PeakFit4.12对色谱峰的峰高、峰宽、保留体积、对称因子各参数进行比较。选择一种分离纯化效果较好的离子交换色谱类型,使用上样浓度为1mg/mL(上样体积50mL),分别采用0~1mol/LNaCl、CH3COONa、NH4Cl溶液为洗脱剂,洗脱流速为1.0mL/min,比较三者洗脱效果,进而确定其最佳洗脱剂和洗脱浓度。结果表明,OH-型离子交换色谱对发菜多糖有更好的分离纯化效果,NaCl的洗脱效果更好,最佳洗脱浓度为0.2mol/L。The isolation and purification of polysaccharide from Nostoc flagelliforme by ion-exchange chromatography with DEAE cellulose were studied. The column of ion-exchange chromatography(Ф2.6×30)was packed with DEAE cellulose anion ion-exchange resin and gradient eluted with NaCl solutions. The content of the polysaccharide was determined with the phenol-sulfuric acid method. The effects of different types of DEAE cellulose resin on the separation efficiency were examined with 0.0~1.00mol/L of gradient elution. PeakFit 4.12 software was employed to collect the chromatographic peak data such as the height,width,retention volume and symmetry factor of the peaks. The elution velocity was 1.0mL/min. The sampling rent elution regents such as NaCl,CH3COONa and NH4Cl on separation efficiency were examined. It was found that the best separation efficiency could be obtained using OH^-type of DEAE cellulose resin with NaCl as the elution regent. With this regent,the comparison of gradient elution with equivalent elution was conducted and the results showed that the separation efficiency of the equivalent elution was better than that of the gradient elution. The optimal elution concentration was 0.2mol/L NaCl. The polysaccharide was further purified with gel chromatography.
