摘  要:目的:研究酸模叶蓼提取物对α-葡萄糖苷酶活性的影响。方法:采用索氏提取法提取酸模叶蓼地上部分,以96微孔板法测定各提取物对α-葡萄糖苷酶的抑制作用,并采用双倒数作图法研究酸模叶蓼甲醇提取物的酶抑制动力学特性。结果:酸模叶蓼提取物具有较好的α-葡萄糖苷酶抑制作用。酸模叶蓼甲醇提取物的抑制效果最好(IC50=2.13μg·mL-1),其次为乙酸乙酯提取物(IC50=3.826μg·mL-1),石油醚提取物抑制活性最小(IC50=230.86μg·mL-1),但均远远大于阳性对照Acarbose(IC50=1103.01μg·mL-1)的抑制活性。抑制动力学实验表明酸模叶蓼甲醇提取物的抑制类型为非竞争性抑制。结论:酸模叶蓼各提取物对α-葡萄糖苷酶活性的抑制效果均很好,具有良好的潜在开发价值。OBJECTIVE:To investigate the effect of the extracts of Polygonum lapathifolium on the activity of α-glucosidase.METHODS:Soxhlet extraction was applied to extract overground part of P.lapathifolium,and the inhibition effect of extracts on α-glucosidase was determined with 96-microplate-based method,and the inhibitory kinetic characteristics of methanol extract was investigated using Lineweaver-Burk method.RESULTS:The extracts from P.lapathifolium showed good inhibitory activity.The methanol extract had the highest inhibitory activity (IC50=2.13 μg·mL^-1)followed by ethyl acetate extract (IC50=3.826 μg·mL^-1),petroleum ether extract (IC50=230.86 μg · mL^-1).And they were all higher than that of acarbose (IC50=1 103.01 μg·mL^-1) as positive control.Inhibitory kinetics test indicated that methanol extract was noncompetitive inhibitor.CONCLUSION:The extracts of P.lapathifolium have good inhibition effect on the activity of α-glucosidase with a good prospect of development and utilization.
