摘 要:建立反相高效液相色谱法检测食品添加剂乳铁蛋白的方法。样品采用氯化钠溶液(0.5mol/L)溶解,过滤后供液相色谱仪测定。采用CAPCELL SG300 C18色谱柱分离,氯化钠溶液(0.5mol/L)、乙腈及三氟乙酸为流动相,梯度洗脱,二极管阵列检测器280nm检测,保留时间及特征光谱图定性,外标法峰面积定量测定。标准曲线线性范围10.0mg/L~100mg/L,相关系数均在0.999以上,定量检测低限5mg/kg。A determination of food additive lactoferrin by reverse phase high-performance liquid chromatography is described.The lactoferrin is extracted from the samples with the sodium chloride solution of 0.5mol/L,filtered,and analyzed at 280 nm by high-performance liquid chromatography with DAD.The chromatographic separation is carried out on CAPCELL SG300 C18 column with ramp mobile phase using the sodium chloride solution of 0.5mol/L,acetonitrile and trifluoroacetic acid.The qualitation of lactoferrin depends on the RT and characteristic UV spectra;quantified by external standard calibration curves.The calibration curve is linear in the 10.0mg/L-100mg/L range with correlation coefficients≥0.999.The limit of detection and quantification is 5mg/kg.The analytical method is simple,fast and precise with a good sensitivity,an excellent reproducibility.
