摘 要:[目的]观察向土壤中加入外源纤维素酶后土壤原生纤维素酶活性及其底物酶解率的变化。[方法]以玉米秸秆为底物,在pH值6.5,温度27、37、47℃的环境中,向土壤施加外源纤维素酶,研究不同温度下外源纤维素酶对土壤原生酶活性及其底物酶解率的影响。[结果]在pH值6.5环境中,各处理酶活均是先上升后下降的,可能是因为土壤与酶共存状态下酶解速率很快,在培养的前半段时间就将大部分秸秆酶解,在此后几天秸秆残留越来越少,酶活也就逐渐降低。各温度处理下的土壤酶活为47℃>37℃>27℃,且各温度环境下添加外源纤维素酶都有助于提高土壤原生纤维素酶活性。[结论]添加外源纤维素酶有助于提高土壤原生纤维素酶活性。[Objective] The activity of the native cellulase in soil and the variation of its enzymatic hydrolysis to the substrate was measured after the exogenous cellulase was added in the soil.[Method] The maize being taken as the reaction substrate,the effect of the exogenous cellulase added in soil on the activity of native enzyme and its enzymatic hydrolysis to the substrate was explored under the environmental condition of the pH value of 6.5,the temperature of 27 ℃,37 ℃ or 47 ℃.[Results] Under the environmental condition of the pH value of 6.5,the activity of enzyme was firstly raised and then decreased,which was possible that the speed of enzymatic hydrolysis could be fasted under the coexist of soil and enzyme,thus,most of straws had been hydrolyzed in early period and in the later,few remains of straws resulted in the reduction of the speed of enzymatic hydrolysis.The order of the activity of enzyme under the temperature was 47 ℃37 ℃27 ℃ and under the treatment of temperature,the activity of native enzyme in soil could be promoted due to the addition of the exogenous cellulase.[Conclusion] The addition of the exogenous cellulose was beneficial to the improvement of the activity of native enzyme in soil.
