摘  要:利用松仁蛋白为原料,以水解度为指标,对Alcalase蛋白酶、中性蛋白酶和胰蛋白酶进行筛选,采用正交试验设计优化酶解条件,制备松仁蛋白多肽,并对多肽进行了组分分析。试验结果表明:Alcalase碱性蛋白酶作为松仁蛋白的水解酶,具有较高的水解度,最佳酶解条件为:温度50℃、pH值8.0、时间180min、酶用量5000U、底物质量分数6%、松仁蛋白酶解的水解度为30.06%。松仁多肽经SephadexG-25凝胶过滤层析分离,得到3个主要组分:PNP1~2的分子质量>23105u、PNP3的分子质量为3776u左右、PNP4~7的分子质量<249u;氨基酸组成分析表明,松仁多肽谷氨酸、精氨酸质量分数较高。The aim of this study was to prepare polypeptide from pine kernel and to analyse its composition.In order to gain a higher degree of hydrolysis,pine kernel protein was hydrolyzed by optimizing enzymes from Alcalase,neutral protease and trypsin according to orthogonal experimental design.Results showed that Alcalase was identified as the best hydrolyzing enzyme.The optimal extraction conditions were obtained as temperature 50 degrees C,pH 8.0,extraction time 180 min,enzyme amount 5000U,and substrate concentration 6%.Under the optimal conditions,degree of hydrolysis was 30.06%.Three components were eluted in the separation by Sephadex G-25,and the molecular mass distribution of each fraction was as follows:greater than 23 105 Da,3 776 Da and less than 249 Da,respectively.Amino acid composition showed that the contents of glutamic acid and arginine were higher.
