摘　　要：目的探讨大蒜素联合生存素反义寡核苷酸(ASODN)对人胃癌细胞系SGC7901体外生长的影响。方法大蒜素联合生存素ASODN处理胃癌细胞系SGC7901,MTT法检测SGC7901的生长;流式细胞仪检测细胞凋亡率;免疫细胞化学法检测Ki67的表达。结果大蒜素联合生存素ASODN用药对SGC7901生长抑制率明显高于大蒜素和生存素ASODN单药组(P<0.05),联合指数为0.46。大蒜素联合生存素ASODN作用于SGC7901细胞12、24和48 h后,细胞凋亡率分别为7.30%、10.44%和16.44%,明显高于单药组(P<0.05)。与大蒜素组、生存素ASODN组和对照组比较,大蒜素联合生存素ASODN作用48 h后,Ki67的阳性表达率下降(P<0.05)。结论大蒜素联合生存素ASODN在抑制胃癌细胞SGC7901的生长和诱导细胞凋亡方面具有协同作用。Objective To investigate the effects of allicin and survivin antisense oligonucleotide（ASODN） on proliferation and apoptosis of gastric cancer cell line SGC7901.Methods The SGC7901 cells were treated by allicin alone or in combination survivin ASODN.After 12,24,48 h,MTT assay was used to examine the inhibition rate of cell growth.The apoptosis of SGC7901 was examined by flow cytometry（FCM）.The expression of Ki67 was detected in gastric cancer cells by immunohistochemical staining.Results Allicin or survivin ASODN effectively inhibited the growth of SGC7901 gastric cancer cells in a concentration-dependent manner at certain range of concentrations.The inhibitory rate of combined treatment with allicin and survivin ASODN was significantly higher than that of allicin or survivin ASODN alone（P0.05） and the value of combination index（CI） was 0.46.In the allicin and survivin ASODN combined treatment group,the apoptosis rate was 7.30%,10.44% and 16.44%,respectively,which was significantly higher as compared with single drug treatment group（P0.05）.The expression of Ki67 protein was down-regulated by allicin plus survivin ASODN compared with single drug treatment and control group（P0.05）.Conclusion Allicin combined with survivin ASODN has synergistic effects in inhibiting the growth and inducing apoptosis of gastric cancer cells SGC7901.
