摘 要:建立了反相高效液相色谱测定陕西稠酒中有机酸类化合物的方法。色谱分析条件:Agilent HC-C18柱(250mm×4.6mm,5μm);流动相为2%的甲醇-0.01mol/L磷酸氢二铵,用1mol/L磷酸调节pH至2.8;流速1mL/min;检测器波长210nm;柱温30℃。面积外标法定量。分析测定了稠酒中8种有机酸,并对4种稠酒样品中有机酸种类和含量进行对比,各成分的线性相关系数除酒石酸为0.9897,富马酸为0.9894外,其余均大于0.99。方法的回收率在96.7%~100.2%之间,RSD为0.52%~3.48%。该方法有快速、分辨率高、重复性好等优点,可用于发酵稠酒中有机酸类化合物的检测。RP-HPLC(reversed-phase high performance liquid chromatography) method for the determination of organic acid compounds in thick liquor was developed. The relative technical conditions were summed up as follows: Agilent HC-C18 chromatography column(250 mm×4.6 mm, 5 μm), mobile phase is 2% methanol- 0.01mol/L diammonium phosphate, with 1 mol/L phosphoric acid adjusted pH to 2.8, flow rate was 1mL/ min,determination wavelength was 210 nm, column temperature was 30 %. Analysis and determination of eight organic acids in four thick liquor samples, and compare the types and content of these organic acids. Linear correlation coefficient were above 0.99, except for tartaric acid was 0.9897 and fumaric acid was 0.9894. The recoveries were between 96.7%-100.2%, RSD were between 0.52%-3.48%. The method was distinguished for its high rapid, high resolution, good reproducibility. It can be applied to assay organic acid compounds in fermented thick liquor.
