摘 要:建立了乳品中大肠菌群检测片快速检测方法,即以国标法中的乳糖发酵管培养基为基本营养成分,优化添加辅助营养成分、指示剂、缓冲剂、选择性抑菌剂,通过冷水凝胶剂等均匀固化于基片上,样品滴加检测片上,培养16~18 h即可直接依据检测片颜色的变化定量检样中大肠菌群的含量,结果可用MPN值报告。实验以多管发酵法为对照,分别用标定菌浓的大肠杆菌菌液和市售乳品样进行对比检测,结果表明,检测片的最低检出限为0~4 mL-1,所测结果与国标法结果相比符合率为96.7%,样品检测报告时间缩短50多小时,两种方法所测结果在统计学上无显著性差异(P>0.05),快速检测片作为一种价廉、简单、快捷、准确的检测方法,能应用于乳品中大肠菌群的快速检测。In this study,a rapid and simple method for detection the total coliforms in dairy,fixing optimal nutrient including basic nutrition and supplementary nutrition,indicating agent,buffering agent,specific bacteriostatic agent on the designed strip through cold gel was investigated.Based on strip color change,coliforms can be quantitatively reported as MPN,and the overall assay time only needed of 16-18 hours.The conditions of the detection such as the species of cold gel and proportion of medium,as well as the concentration of indicating agent and buffering agent were studied.This method was compared to the multiple-tube fermentation using calibration of standard strain for the analysis of samples,the rapid test strip showed low detection limit of 0-4 cfu/mL.A high agreement(96.7%) was found between rapid test strip and standard on 60 commercial dairy samples.No significant difference(p0.05) was found between the detection results for the methods.The test strip method was cost-effective,simple,rapid and accurate,can be used to detect coliforms in dairy.
