摘  要:对一株畜禽用凝结芽孢杆菌菌株进行了发酵条件的优化,通过单因素实验对培养条件、培养基成分进行了筛选,通过正交实验对培养基组成进行了优化。结果表明,该菌株的最佳发酵条件:温度35℃、转速220 r/min、装液量10%、接种量3%、初始pH=7.0、发酵时间42 h;最佳培养基组成:葡萄糖8.0 g/L,酵母膏12.5 g/L,KH2PO4 3.0 g/L,MnSO4.H2O 0.18 g/L,MgSO4.7H2O0.125 g/L。在此最佳发酵条件和培养基组成下,发酵液活菌数达38×108 cfu/mL。The fermentation conditions of bacilltus cougulans were optimized, in which the culture conditions and ingredients of culture medium were screened through single factor experiment, and the ingredients of culture medium were optimized by orthogonal experiment. The results showed that the optimal culture conditions were: temperature 35℃, rotational speed 220 r/rain, liquid volume 10%, inoculation amount 3%, initial pH=7.0, fermentation time 42 h; the optimal ingredients of euhure medimn were: glucose 8 g/L, yeast extract 12.5 g/L, KH2P04 3.0 g/L, MnSO4·H20 0.18g/L, MgSO4·7H20 0.125 g/L. Under the optimized t;ermentation conditions and in the optimized ingredients of cuhure medium, the living bacteria amount can reach 38×108cfu/mL.
