摘 要:利用NCBI公共数据库下载获得的3 306条茶树EST,去除其中低质量和冗余的序列,得到全长为635.46 kb的1 335条无冗余EST,在这些序列中共发现216个SSR,分布于185个EST中,出现的频率为16.18%,平均每2.94 kb的表达序列中有1个SSR。茶树的EST-SSR类型丰富,其中二核苷酸重复出现的频率最高,占主导地位。利用Primer 5.0软件从185条含有SSR的EST中设计引物100对,从中随机选取60对引物检测EST-SSR在云南茶树资源中的多态性。结果表明,有30对引物在供试的63份茶树资源中得到有效扩增,并具有多态性;共检测到等位基因数71个,每对引物检测出的等位基因数2~8个,平均4.9个;多态信息量(PIC)变幅为0~0.731,平均为0.499;观察杂合度变幅为0~0.970,平均0.392。EST-SSR标记分析表明云南茶树资源具有较高的遗传多样性。The frequency and characteristics of SSR were analyzed and clarified in ESTs of tea plant(Camellia sinensis(L.) O.Kuntze).3 306 ESTs of tea plant were downloaded from the public database of NCBI,among which the redundant or low quality were removed,finally obtained 1335 non-redundant ESTs with total length of 635.46 kb.Totally 216 SSRs distributed in 185 ESTs were found,accounting for 16.18% of the non-redundant ESTs,and the average distance of distribution is 1/2.94 kb.EST-SSR types in tea plant are considerably abundant,among which the dinucleotide repeat is the dominant type.These results provide a base for the development and application of EST-SSR markers in tea plants.According to tea ESTs,60 SSR primer pairs form 100 tea EST-SSRs were designed with Primer 5.0 software.After tested on the annealing temperature and polymorphism of primers,30 of them produced stable and clear bands with better polymorphism in 33 tea germplasms.A total of 71 putative alleles were generated,and ranged from 2 to 8,with a mean of 4.9 for putative alleles per locus.The polymorphism information content(PIC) varied from 0.00 to 0.731and the observed heterozygosity(Ho) ranged from 0.00 to 0.970,with an average of 0.499 and 0.392,respectively.All the tested SSR primers showed a high level of genetic polymorphism.
