摘  要:在单因素试验的基础上,通过Box-Benhnken组合设计和响应曲面分析法优化香菇多糖径向流色谱分离的工艺条件。响应面分析结果表明,当样品浓度为10mg/mL,上样量31mL,上样流速19mL/min,洗脱流速27mL/min,在次条件下多糖回收率达到87.33%,比传统轴向色谱分离及化学方法效率更高,时间更短。Radial flow chromatography(RFC) packed with ion-exchange resin was used to purify the Lentinula edodes crude polysaccharides.Based on the single factors of purification efficiency,response surface experiments(RSM) was employed to obtain the optimization.The purification efficiency of RFC was compared with that of axial column and traditional chemical method.RSM analysis showed good correspondence between experimental and predicted values.The optimal conditions for the purification of Lentinula edodes polysaccharides with RFC were determined: sample concentration of 10mg/mL,sample volume of 31mL,sample flow-rate of 19mL/min and elution flow-rate of 27mL/min.The recovery of polysaccharide is 87.33% under these conditions.The comparative test results indicated that the best purification efficiency was obtained by RFC within the shortest time.
