摘 要:在摇瓶和发酵罐中研究了添加正十二烷对三孢布拉霉菌合成β-胡萝卜素以及浸提色素效果的影响。结果表明,在摇瓶和发酵罐中(200r/min和350r/min转速)添加5%的正十二烷能够将20.3mg/L、53.2mg/L和52.9mg/L的β-胡萝卜素浸提到有机溶剂中(分别占胞色素内含量的4.0%、7.0%和6.8%),同时添加正十二烷提高了发酵罐中的溶氧水平(kL)a,当搅拌转速为200r/min和350r/min时,与对照相比,胞内β-胡萝卜素的合成水平提高了10.8%和4.4%,并且缩短了发酵周期,使β-胡萝卜素的生产效率提高了24.7%和25.4%。以上结果表明,正十二烷可以同时作为浸提剂和氧载体提高三孢布拉霉菌合成β-胡萝卜素的水平,并在较低的搅拌转速条件下可获得较高转速下的发酵水平,降低了生产成本。The effects of dodecane on production and extraction of β-carotene produced by Blakeslea trispora were investigated in shaking flask and fermentor. The results showed that 20.3mg/L, 53.2mg/L and 52.9mg/L β-carotene were extracted by 5% dodecane into the organic phase in shaking flask and fermentor with rotational speed of 200r/min and 350r/min, which accounted for 4.0%, 7.0% and 6.8% of intracellular β-carotene content, respectively. In addition, the intracellular β-carotene production and productivity were enhanced by dodecane due to the improvement of dissolved oxygen concentration indicated by the increment of kLa. Compared with control, the yield of intracellular β-carotene was improved by 10.8% or 4.4%, while the productivity was improved by 24.7% or 25.4%, when the rotational speed was 200 r/min or 350 r/min respectively. These results suggested that dodecane can be simultaneously used as extraction solvent and oxygen-vector to improve the β-carotene production by B. tr/spora. The level of β-carotene observed at high agitation speed can be obtained at low agitation speed, which can reduce the production cost, and has a good potential for industrial application.
