摘  要:采用高速逆流色谱法分离花椒中的生物碱。逆流色谱条件:正己烷-乙酸乙酯-甲醇-水(5∶5∶7∶5)为溶剂体系,上相为固定相,下相为流动相,流速2mL/min,仪器转速800r/min,检测波长254nm,上样量800mg。在此条件下,一次分离得到组分A(130mg)和组分B(15mg),经薄层色谱定性分析为生物碱,HPLC测定组分A和B的纯度分别为92.8%、85.4%。High- speed counter- current chromatography (HSCCC) with a two- phase solvent system composed of n- hexane- ethyl acetate- methanol- water ( 5:5 :7 :7, v/v) was applied to the isolation and purification of alkaloids from Zanthoxylum bungeanum Maxim.The lower phase was used as the mobile phase and performed at a flow rate of 2mL/min,while the apparatus rotated at 800r/min, and detected at 254nm.Two kinds of alkaloids were obtained and yielded 130mg(A),15mg(B)from 800mg of crude extract in a one-step separation.A and B were identified by Thin Layer chromatography(TLC) ,and the results showed that the two fractions were alkaloids,with the purity of 92.8% and 85.4% ,determined by high performance liquid chromatography(HPLC).
