摘 要:以福尔马林灭活的鼠伤寒沙门氏菌免疫雌性日本大耳兔,制备抗沙门氏菌的多克隆抗体。分别通过辛酸-硫酸铵沉淀法、磁珠偶联抗原吸附法纯化抗体,所制备的抗体效价为3.2×105,纯度较高,但交叉反应现象较严重。本实验以杂菌抗原反向吸附法纯化抗体,有效消除了抗体与杂菌的交叉反应,初步建立了一种高特异性多克隆抗体的纯化方法。A highly specific anti-Salmonella typhimurium polyclonal antibody was prepared for immunological determination of Salmonella typhimurium. Anti-Salmonella typhimurium polyclonal antibody was generated in Japanese rabbits using forrnalin- inactivated Salmonella typhimurium as antigen and purified by octanoic acid-ammonium sulfate precipitation and magnetic beadantigen coupling. The titer of the prepared highly pure antibody was 3.2 × 10^5; however, this antibody revealed serious crossreaction with other bacteria. In this paper, the antibody was purified by miscellaneous bacterial antigens in a reverse adsorption mode, which could eliminate the cross reaction between the antibody and miscellaneous bacteria. Therefore, an effective purification method to obtain specific polyclonal antibody was established.
