摘 要:[目的]筛选对花椒籽油具有偏好性的脂肪酶产生菌。[方法]采用甘油法和气相色谱相结合的方法,从实验室分离到的11株具有甲醇耐受性脂肪酶菌株中筛选对花椒籽油具有转酯活性的菌株,并进行了最佳产酶条件的优化。[结果]11株菌株中以D-1-4菌脂肪酶催化花椒籽油的转酯率最高,达到43.05%;其最佳产酶条件为:以2.5%蔗糖为碳源、2.0%黄豆粉+1.0%玉米粉为复合氮源、1.0%橄榄油为诱导物,在初始pH 6.0、30℃培养48h时脂肪酶活力达到79.30U/ml。[结论]为脂肪酶产生菌及其脂肪酶的应用提供了依据。[Objective] The strains with transesterification activity to Chinese prickly ash seed oil were screened.[Method] Strains with transesterification activity were screened from 11 methyl-resistant lipase-producing strains,which have been isolated by previous research in our laboratory.And the optimum enzyme producing conditions were optimized.[Result]In 11 strains,the conversions of Chinese prickly ash seed oil catalyzed by D-1-4 lipase was up to 43.05%,and the optimum culture conditions were as follows:2.5% sucrose as carbon source,2.0% soy bean power + 1.0% corn power as compound nitrogen sources,adding 1.0% olive oil to the culture medium,initial pH 6.0,incubated at 30 ℃ for 24 h,the lipase activity reached 79.30 U/ml.[Conclusion]All those provided basis for the application of lipase and lipaseproducing strains.
