摘 要:[目的]探讨废次烟叶中茄尼醇的提取纯化工艺。[方法]采用超声波辅助提取技术,结合柱色谱法对废次烟草中茄尼醇的提取、分离和纯化进行了研究。[结果]通过正交试验获得茄尼醇最佳提取条件为:超声功率120W,超声时间60min,料液比1:10,提取温度70℃。提取液经皂化、酸化后,用正己烷萃取得到纯度为21.5%的茄尼醇粗品;粗品经重结晶后得到纯度为77.6%的茄尼醇产品;进一步通过柱色谱分离,石油醚-乙酸乙酯(8/1,V/V)洗脱,收集相关流分,结晶得到纯度为99.2%的茄尼醇产品,整个工艺过程中茄尼醇产品的得率为0.512%。[结论]应用超声波辅助提取废次烟叶中的茄尼醇,具有萃取速度快、效率高的优点,适合工业化生产。[Objective]The aim was to study isolation and purification technology of solanesol from discarded tobacco.[Method]The extraction,isolation and purification technology of solanesol from discarded tobacco were studied by adopting ultrasonic-assisted extraction technique and column chromatography.[Result]The optimum extraction conditions of solanesol from discarded tobacco were obtained through orthogonal experiments as follows:the u1trasonic power was 120 W;the extraction time was 60 min;the proportion of raw materials and solution was 1:10;and the extraction temperature was 70 ℃.Then solansol was extracted from solansol extract by hexane with purity of 21.5% after saponification and acidification;after that the solansol with 77.6% purity was obtained by recrystallization;finally the crude solansol was purified on silica column chromatography using petroleun ether/ethyl acetate(8 /1,V/V) as mobile phase.The purity of solanesol was 99.2%,and the yield of solanesol was 0.512%.[Conclusion]Ultrasonic-assisted extracting solanesol from discarded tobacco has advantages of fast speed and high efficiency,so it is suitable for industrial production.
