耐高温酒精酵母诱变育种研究

摘　　要：以嗜鞣管囊酵母P-01为出发菌株,经亚硝基胍诱变处理,采用TTC筛选平板初筛,杜氏管发酵二级筛选和混合糖发酵三级筛选,得到3株38℃发酵温度时的乙醇高产菌株P-01-33、P-01-70和P-01-190,其乙醇产率为12.9g/L、12.1g/L和13.3g/L,分别比出发菌株P-01的乙醇产率(9.5g/L)提高35.80%、27.47%、30.81%。Pachysolen tannophilus P-01 was used as starting strain for breeding thermostable alcohol producing yeast with nitrosoguanidine（NTG） mutation.With the ordering screening with TTC plate,Durham tube fermentation and fermentation with mixed sugar（hexose and pentose）,three mutant stains named as P-01-33,P-01-70 and P-01-190 were isolated.Yield of alcohol reached 12.9 g/L,12.1 g/L and 13.3 g/L respect for the three strains at 38℃,which were 35.80%,27.47% and 30.81% higher than that of the starting strain P-01（9.5g/L）,respectively.
