摘　　要：目的:研究我国汉族人群CCL3L1基因拷贝数的分布,及其与非静脉吸毒者慢性乙肝的相关关系。方法:选1 477例慢性乙肝病人及1 023例健康人。用qPCR方法检测CCL3L1基因的拷贝数;用qPCR及ELISA方法分别检测CCL3L1基因拷贝数与其mRNA、蛋白质的定量关系,并用Transwell法分析了不同拷贝数目与其趋化效力的关系。用Pearson's卡方检验和Logistic回归方法检测CCL3L1基因拷贝数目变异与乙肝易感性的统计学相关性。结果:CCL3L1基因拷贝数目在我国汉族正常人为0-9(中位数=3),与其mRNA、蛋白质产物呈正相关,也与其趋化性呈正相关关系。低拷贝数目者比健康对照更易患乙肝(P<0.01)。结论:在中国汉族人群中,CCL3L1基因拷贝数目变异与乙型病毒性肝炎易感性存在显著相关,低拷贝数目者更容易患乙肝。AIM：To determine the distribution of CCL3L1 copy numbers in Chinese Han population,and investigate the correlation between the variation of CCL3L1 copy number and chronic hepatitis B in the patients who are non-drug abusers.METHODS： A case-control（1 477 cases vs 1 023 controls） approach was undertaken.The copy numbers of CCL3L1 were quantified by qPCR.The correlation of CCL3L1 copy number with its mRNA expression was measured by qPCR and the protein level of CCL3L1 was detected by ELISA.The chemotactic effects of different copy numbers of the gene were analyzed by Transwell assay.Pearson＇s chi-square test and Logistic regression were used to examine the association between the copy number of CCL3L1 and hepatitis B.RESULTS： The distribution of the copy number among Chinese Han peoples ranged from 0-9（median=3）.CCL3L1 copy number was concordant with its mRNA expression and protein products,as well as its chemotactic effects.The copy numbers of CCL3L1 in hepatitis B patients were lower than those in the healthy controls（P〈0.01）.CONCLUSION： The copy number of CCL3L1 is a heritable factor associated with susceptibility to chronic hepatitis B among Chinese Han population.Low copy number of CCL3L1 indicates higher risk of hepatitis B carriage.
