摘 要:目的:建立难溶性食品中添加剂定量测定方法。方法:样品加入新型LHJ增溶剂,使其中的食品添加剂充分溶解,加入亚铁氰化钾、乙酸锌沉淀蛋白,定容,离心,取上清液过0.45μm滤膜,取10μl进高效液相色谱仪测定。采用C18色谱柱,以甲醇+乙酸铵(5+95)为流动相,流速1.0 ml/min,柱温30℃,波长230 nm紫外检测器,根据保留时间定性,外标峰面积定量。结果:该方法在0.010 mg/ml~0.100 mg/ml范围内线性关系良好,相关系数均大于0.9999,检出限为0.00015 g/kg回收率在90%~108%之间。结论:该方法简便,快捷,回收率高,精密度好,处理样品时使用新型LHJ增溶剂测定结果提高2倍~3倍。Objective:To establish a method for quantitative determination of additives in insoluble food.Methods: The new LHJ solubilizer was added into samples to make the food additives fully dissolved.Then potassium ferrocyanide and zinc acetate were added to precipitate protein.After constant volume and centrifugation,the supernatant was filtered over 0.45 μm membrane,then 10 μl filtrate was taken for determination by high performance liquid chromatography.C18 column was used with methanol and ammonium acetate(5+95) as the mobile phase,flow rate of 1.0 ml/min,column temperature at 30℃ and UV detector wavelength at 230 nm in this method,then qualitative and quantitative analysis were conducted according to retention time and peak area respectively.Results: Several additives in 0.010 mg/ml^0.100 mg/ml are in a good linear relationship,the correlation coefficients are greater than 0.9999,the detection limit is 0.00015 g/kg,the recovery rate is in the range of 90%~108%.Conclusion: The method is simple and rapid with high recovery rate and good precision.Using the novel LHJ solubilizer in sample preparation can improve efficiency of results by 2~3 times.
