摘 要:以玉米秸秆粉为主要原料,在单因素实验基础上利用正交实验对航天诱变高产纤维素酶菌株黑曲霉ZM-8产β-葡萄糖苷酶的培养基组分进行了优化。研究结果表明,麸皮添加量(F=32.414>F0.01=6.01)和含氮量(F=13.716>F0.01=6.01)对该菌株产酶的影响达到极显著水平,含水量达到了显著水平(F=4.251>F0.05=3.55)。该菌株产酶的最佳培养基组分为:麸皮添加量为35%,氮源为1.2%的(NH4)3PO4,含水量为250%,接种量为1∶18。在此条件下,β-葡萄糖苷酶活为42.49U/mL。Abstract:Taking corn stalk powder as the main raw material, the medium components of the aerospace mutant Aspergillus niger ZM-8 with high cellulase activity for producing LS-glucosidase by the orthogonal experiment was optimized based on the single factor experiment.The results showed that the effects of bran content( F = 32.414 〉 F0.01 =6.01 ) and nitrogen content( F = 13.716 〉 F0.01 = 6.01 ) of the strain producing 15-glucosidase were extremely significant level,the effect of water content was significant level (F = 4.251 〉 F0.05 = 3.55).The optimization medium components of the strain producing β-glucosidase was as follows.the bran content was 35% ,the nitrogen source was 1.2% of (NH4)3PO4, the water content was 250% and the inoculation amount was 1:18. Under these conditions,the 13-glucosidase activity reached 42.49U/ mL.
