摘 要:黑曲霉ZM-8是一株经航天诱变筛选出的高产纤维素酶菌株。以玉米秸秆和麸皮为原料,利用固态发酵法研究了发酵时间、发酵温度和初始pH对该菌株产β-葡萄糖苷酶的影响,采用正交试验对各因素进行了优化,并对酶学特性进行了初步研究。结果表明,发酵时间和发酵温度对酶活影响均达到极显著水平(F=14.994>F0.01=5.85;F=10.872),初始pH(F=5.843)对其影响达到显著水平;当初始pH为5.5、培养温度为35℃、培养时间为72 h时,β-葡萄糖苷酶的酶活达到了58.95 U/mL。该酶最适温度为55℃,保温2 h后仍具有95%以上的酶活力;最适pH为5.5,pH在3.0~6.0时,酶活力稳定性良好。The strain Aspergillus niger ZM-8 with high cellulase activity was screened by space mutation with the corn stalk and bran as raw materials, the effects of fermentation time,fermentation temperature and initial pH on the β- glycosidase activity were studied by the method of solid-state fermentation and these factors were optimized by the orthog- onal experiment, and the enzymatic characteristics were determined in this paper.The results showed that: the effects of fer- mentation time(F=14.994)and fermentation temperature (F=10.872) on the β- glycosidase activity were extremely signifi- cant, the effect of initial pH was significantly (F=5.843〉Fo.=3.49).The β-glycosidase activity reached 58.95 U/mL under the conditions of initial pH was 5.5,fermentation temperature was 35 ℃ and fermentation time was 72 h.The β-glycosi- dase optimun temperature was 55℃ and the residual enzymatic activity was more than 95 % after being kept for 2 h.lts optimun pH was 5.5 and the enzyme activity was stable from pH 3.0 to 6.0.
