摘  要:豆粕、菜粕按1:1混合为发酵基质,经过菌株初筛、复筛及单菌、混菌的固态发酵,以酸溶性蛋白含量(TCA-N)、蛋白水解度(DH)以及酶切位点有效值(EV)、硫苷降解率为主要评价指标,得到菌株的最佳组合为BL-1+BS+解脂假丝酵母。通过4因素3水平正交试验,确立的最佳发酵工艺条件为料水比1∶1.1、接种量9%、初始发酵温度37℃、发酵时间48 h。在此条件下发酵后样品粗蛋白含量从49.5%增加到53.1%,TCA-N由2.50%升高到27.2%,硫苷降解率达65.2%。SDS-PAGE图谱显示大分子蛋白被降解为12 ku以下。The solid-state fermentation mixed with rapeseed meal and soybean meal by 1:1 was studied. After strains screened, rescreened and solid-state fermentation by single stain, mixed stains, the best combination (BL-l+BS+Candida lipolytica) was obtained. The degradation rate of glucosinolate was used as the main evaluation indexs, as well as the TCA-N content, the degree of protein hydrolysis (DH), the effective value of enzyme cutting site (EV). The result of the orthogonal experiment of four factors and three levels showed that the optimal fermentation conditions were ratio of material to water 1:1.1, initial fermentation temperature 37℃, mixture inoculum 9%, fermentation time 48 h. Under this conditions, the TCA-N content rised from 2.50% to 27.2%, the degradation rate of glucosinolate reached 65.2%. The results of SDS-PAGE showed that the larger molecular proteins were degraded into the smallers lower than 12 ku.
