摘 要:为了缩短大肠菌群的检测时间,满足蔬菜、水果等易腐食品的检测需要,建立了一种利用分光光度法快速、准确检测大肠菌群数的新方法。该方法建立在传统的大肠菌群检测方法的基础上,对传统多管发酵法进行改进,将制得的伊红美兰混合液加入到乳糖胆盐培养基中作指示剂,通过其产生沉淀后溶液颜色的变化来指示大肠菌群的数量,2.5h即可得到检测结果。结果表明,向1mL乳糖胆盐培养基中加入60μL伊红美兰溶液时指示效果最好;在37℃培养2.5h后显色稳定,检测结果重现性好,精密度相对标准偏差小于2.75%。In order to reduce the detection time of total coliforms counts and satisfy the detection requirements of the corruptible foods,such as vegetables and fruits,a rapid and accurate enumeration method of total coliforms counts based on spectrophotometry method was developed. This rapid method was based on the traditional multiple tube fermentation method,but some manipulation steps had to be improved. The eosin methylene- blue solution was added into lactose bile salt medium to act as the indicator. The degree of color change of the solution can indicate the total coliforms counts. The detection time was 2.5h. The results indicated that the best indicate effect could be obtained when 60μL eosin methylene-blue solution was added into lmL lactose bile salt medium. After cultured it for 2.5h at 37℃,the displayed color was stable. The reproducibility of the detection results was well and the precision(RSD) was less than 2.75%.
