摘 要:根据植物叶绿体rbcL基因的序列比对情况,在保守区设计植物通用引物,并针对4种油料的变异区设计相应的特异探针,将探针与不同荧光编码的微球进行偶联,偶联后的微球与生物素标记的PCR产物杂交,利用Bio-PlexTM 200的液相芯片平台检测荧光信号,建立了大豆、花生、橄榄和葵花籽4种常见食用油的液相芯片鉴别方法。该方法能同时特异性识别4种食用油,且灵敏度高。A new method for identification of four common edible oils based on suspension array was estab- lished. According to the results of rbcL gene sequence alignment, one pair of universal primers targeting plant conserved region and four species - specific probes targeting the variable region of four oil plants were designed . Each probe was coupled to a set of unique color - coded microspheres. Biotinylated PCR ampli- cons of four edible oils were hybridized to the complementary probes on microsphere sets. Bound amplicons were detected fluorometrically using Bio -PlexTM200 suspension array. This method could specifically iden- tify edible soybean oil ,peanut oil ,olive oil and sunflower seed oil at the same time and had high sensitivity.
