摘  要:大分子量透明质酸高产菌株的选育是工业化生产透明质酸的关键,本实验以兽疫链球菌为出发菌,采用亚硝基甲基脲和15keV低能氮离子进行复合诱变,筛选出一株溶血素和透明质酸分解酶双缺陷型突变菌株SZ-36,采用摇瓶发酵培养后,透明质酸的产量最高达到3.16g/L,相对分子量达到2.42×106u,比原始菌株分别提高了267.4%和137.3%,并经10次传代以后,突变菌株SZ-36具有良好的传代稳定性,为工业化生产透明质酸提供了优良菌株。It was very important for industrialized production of hyaluronic acid to select excellent stain. Streptococcus zooepidemicus was subjected to mutagenesis by means of methylnitrosourea and low-energy ion implantation of 40× 10^14ions/cm^2 dose at 15keV, After culture in selective medium and screened out a mutant strain with nonhemolytic and negative hyaluronidase designated as SZ-36, The yield of hyaluronic acid reached up to 3.16g/L and relative molecular weight of 2.42×10^6u by shake flask culture, increasing by 267.4% and 137.3% respectively than the original strain, The strain still revealed good stability after transferring ten generations, It can provides excellent strains for the industrial production of hyaluronic acid.
