摘  要:目的建立一种前处理简单、灵敏度高、重现性好的馒头中合成色素的检测方法。方法采用高效液相色谱-二极管阵列检测(HPLC-DAD)法,样品用无水乙醇+氨水+水(7+2+1)溶液提取,Agilent HC-C18柱(250mm×4.6mm,5μm)液相色谱柱分离,乙酸铵缓冲溶液(0.02mol/L)-甲醇为流动相进行梯度洗脱,检测波长448nm。结果测定结果具有良好的线性和重复性(RSD:1.25%~2.85%),样品加标回收率为87.4%~90.3%,方法检出限为0.002g/kg。结论该方法具有简便、快速、准确等优点,可用于准确测定馒头中的合成色素。Abstract: Objective To establish a detecting method for determining the synthetic color in steamed bread with simple pretreatment, high sensitivity and good repeatability. Method By the method of high - performance liquid chromatography with diode array detection ( HPLC - DAD), the samples was extracted by the solution of absolute ethyl alcohol, ammonia water and clear water (with the ratio of 7:2:1 ), and separated by liquid chromatographic column of Agilent HC -C18 column (250mm x 4. 6mm, 5pLm) . Conduct gradient elution for the mobile phase by ammonium acetate buffer solution - methanol. The detected wavelength was 448nm. Results The detected result showed good linearity and repeatab (RSD) were between 1.25% and 2.85%. The recovery rate of the and the detection limit of this method was 0. 002g/kg. Conclusions rate, and it was suitable for the determination of synthetic color in ility, and the relative standard deviation samples was between 87.4% and 90. 3%, The method was simple, rapid and accu- steamed bread.
