摘  要:筛选得到的野生米曲霉产高转苷活性β-葡萄糖苷酶,可以通过酶转苷作用合成龙胆低聚糖,通过亚硝基胍和紫外线复合诱变,经过七叶苷平板和发酵测酶活复筛,快速筛选得到一株遗传稳定性好的菌株ALQ1,β-葡萄糖苷酶酶活力达到241.43U/g,较原始菌株提高了88.62%。Abstract: The crude β-glucosidase of a wild Aspergillus oryzae has a high transglycosylation activity, which enables the enzymatic synthesis of gentiooligosaccharides. This wild Aspergillus oryzae was used as an original strain to carry out mutagenetic breeding adopting UV and NTG, respectively. The spore suspension of A. oryzae was treated by 0.1 mol/L NTG for 30 minuites, followed with UV irradiation for 50 seconds, under magnetic stirring. After screening in Esculoside plates and fermentation, a mutant A. oryzae AH1 which was able to produce β-glucosidase with high yield and could be stably passed on genetics was eventually found. The β-glucosidase yield of A. oryzae AH1 was 241.43 U/g, enhanced more than 88.62% higher than that of the original strain.
