Optimization of Solid-state Fermentation Conditions of β-glucosidase Production by Aspergillus niger HQ-1 with Response Surface Methodology

摘  要:采用单因素试验和响应面法对黑曲霉(Aspergillus niger)HQ-1产β-葡萄糖苷酶的固体发酵条件进行了优化,得到产酶的最佳发酵条件为:玉米秸秆粉6.0 g、麦麸6.0 g、(NH4)2SO41.5 g、KH2PO41.6 g、MgSO4.7H2O 0.8 g、含水量73.4%、起始pH3.91、培养温度和培养时间分别为33.7℃和96 h。优化后,β-葡萄糖苷酶比活力最高为8.244μkat/g,比未优化的酶比活力最高值(1.700μkat/g)提高了3.85倍。Single-factor test and Response surface methodology (RSM) were used to optimize solid-state fermentation (SSF) conditions for β-glucosidase production by Aspergillus niger HQ-1. The optimal fermentation conditions were:corn stover powder 6.0 g,wheat bran 6.0 g, (NH4)2SO4 1.5 g,KH2PO4 1.6 g, MgSO4·7H2O 0.8 g, moisture content 73.4 %,initial pH 3.91, cultivation temperature and time were 33.7 ℃ and 96 h, respectively. After optimization, β-glucosidase specific activity was 8.244 μkat/g, 3.85 times higher than the enzyme specific activity (1.700 μkat/g) under the unoptimized conditions.
