摘 要:从天山花楸茎、叶和果实中分离到53株内生细菌,利用刚果红初筛平板获得透明圈较大的4株菌,在此基础上进行摇瓶复筛,得到一株产纤维素酶活力较高的菌株(S11)。该菌发酵培养12h,产羧甲基纤维素酶(CMCase)酶活力最高达到39.47U/mL。该酶最适反应pH值为5.0,在pH 5.0~6.0范围内较稳定;最适反应温度为50℃,在50℃以下酶的热稳定性较好,60~70℃几乎完全失活;各种供试金属离子与化学试剂对酶活性都有不同程度的抑制作用,其中Ca2+、SDS和EDTA的抑制作用最强。Fifty-three endophytic bacteria were isolated from Sorbus tianshanica Rupr.stems,leaves and fruits.Four colonies were surrounded by big transparent zones in congo red agar medium.A strain(S11) with higher cellulase activity was isolated and cultured in shake flasks for secondary screening.Maximum carboxymethyl cellulase(CMCase) activity was found to be 39.47 U/mL after 12 h culture.The optimal reaction pH for crude CMCase solution was 5.0,and it revealed high stability at pH 5.0–6.0.The optimal reaction temperature was 50 ℃,and its activity remained stable below 50 ℃ but was almost completely lost at 60–70 ℃.The CMCase activity was inhibited by all tested metal ions and compounds,and dramatically inhibited by Ca2+,SDS and EDTA.
