摘 要:以壳聚糖为唯一碳源,筛选获得壳聚糖酶活力较高的菌株,根据菌株菌落形态及ITS序列分析,初步鉴定该菌株为烟曲霉(Aspergillusfamigatus)。该壳聚糖酶的最适反应温度为55℃,最适pH值为5.6,在45~60℃和pH值4.0~8.0范围内稳定,Mn2+对酶有明显的促进作用,Ag+、Cu2+、Zn2+对酶有明显的抑制作用,其酶学特征表明该壳聚糖酶在壳聚糖降解方面具有应用潜力。The aim of this paper was to screen chitosanase producing strain and to investigate the characteristics of the chitosanase produced by this strain. Chitsoan was used as the sole carbon source for the acclimatization and screening of strain. A strain with a higher chitosanase activity was isolated from soil. It was identified as Aspergillus fumigatus by its morphology in solid medium and by analyzing the sequence of ITS. The optimum temperature and pH were 55℃ and 5.6, respectively. And the ehitosanase is stable at 45-60℃ and pH 4-.0-8.0, respectively. The activity of the enzyme is strongly promoted by Mn2+ and inhibited by Ag+, Cu2+ and Zn2+. The properties of ehitosanase indicated that it has a great potential application in ehitosan degradation. :
